Development of probiotic strain diagnostics for the control of bacterial diseases in hatchery rearing of aquaculture species



The primary objective of the project is the development and diagnostic characterization of beneficial species of bacteria for protection against disease-causing bacteria encountered in the seed production and grow-out rearing of various aquaculture species (probiotics). The proposed project is anticipated to span three years and will be divided into five (5) phases. In this initial project, probiotics will be developed for use in two species that are produced at ISL, geoducks and oysters, three new species: sablefish, sea urchins and abalone, and if successful, later expanded to cover other species such as salmon. Each of the species of bacteria making up the probiotic mixtures will be genetically characterized to enable the production of consistent high quality mixtures of beneficial bacteria.

Probiotics will be developed from the gut bacteria of six shellfish species (manila clams, geoducks, oysters, urchins, abalone, and scallops) and one fin-fish species (sablefish). These bacterial species in the probiotic mixtures will be identified through comparison of DNA sequences to the large database of bacterial sequences in GenBank.

Common pathogenic bacteria for each shellfish or fish species will be characterized. Control sets of standard pathogenic bacterial DNA sequences will be developed for use in diagnostics and in the measurement of probiotic effects on pathogen concentrations.
